The project involves reviving agro strains from various collections and characterizing the strains for antibiotic resistance. Agro strains that do not show antibiotic resistance to kanamycin, gentamicin and spectinomycin will be sent out for whole genome sequencing and alignment. Genomic data will be collected for potentially disarming the virulence genes. Research will continue by testing the antibiotic sensitive agro strains for the ability to be transformed with a binary vector, to form gall tissue on various plant species/cultivars, and the ability to facilitate transfer the T-DNA from the binary vector introduced. Strains that can produce gall tissue will be evaluated for rate of tissue growth and homogeneity of T-DNA/tissue expression. Time permitting successful agro strains will be disarmed and genome modified with GAANTRY technology. The incumbent will be expected to work independently to conduct research and apply statistical analysis and programming techniques on data collected on citrus and grape cultivars. Department Overview Part of the Department of Plant Pathology in the College of Agriculture and Environmental Sciences, the Coaker Lab focuses on the interaction between bacterial pathogens and plants. Their work focuses on understanding kinase-mediated immune signaling and pathogen effector targets in both model and crop plants. Recent research investigates vascular pathogens, including vector-borne disease associated with Liberibacter species in citrus, tomato and potato. Additional work includes understanding the interaction between the Gram-positive vascular pathogen Clavibacter and its plant hosts. POSITION INFORMATION
